چکیده

Ubiquinol-6 (reduced coenzyme Q6) was more effective than ubiquinone-6 and almost as effective as a-tocopherol in reducing the stable free radical diphenyl-p-picrylhydrazyl. The antioxidant activity of ubiquinol-6 was measured at low oxygen concentrations with an oxygen electrode. Ubiquinol-6 was found to be as e5cient as a-tocopherol as an inhibitor of the heme-catalyzed peroxidation of arachidonic acid emulsions. Ubiquinold was oxidized to ubiquinone-6 by free radical products of the heme-catalyzed decomposition of cumene hydroperoxide. a+Tocopherol did not inhibit this reaction. Light was used to catalyze the peroxidation of mitochondrial lipids at low concentrations of oxygen. This peroxidation was inhibited by the addition of ubiquinol-6 or cY-tocopherol to the mitochondria. Light-induced mitochondrial peroxidation was inhibited by ubiquinol-6 produced from exogenous ubiquinone-6 by the electron transport-linked oxidation of /3-hydroxybutyrate. Ubiquinols may function as antioxidants in the mitochondrion. This could explain why ubiquinones and related compounds have been found effective in the relief of certain vitamin E deficiency syndromes in some species.

Objective: Extracts of Anchomanes difformis (AD) are used in folkloric medicine to treat several diseases and infections. However, their roles in mitochondrial permeability transition pore opening are not known. Material and Methods: The viability of mitochondria isolated from Wistar rat liver used in this experiment, was assessed by monitoring their swel...

Involvement of Cytochrome P-450 in n-Butyl Nitrite-Induced Hepatocyte Cytotoxicity

Addition of n-butyl nitrite to isolated rat hepatocytes caused an immediate glutathione depletion followed by an inhibition of mitochondrial respiration, inhi- bition of glycolysis and ATP depletion. At cytotoxic butyl nitrite concentrations, lipid  peroxidation  occurred  before  the  plasma  membrane  was  disrupted. Cytochrome P-450 inhibitors inhibited peroxynitrite formation and prev...

Biochemical, physiological and medical aspects of ubiquinone function.

This presentation is a brief review of current knowledge concerning some biochemical, physiological and medical aspects of the function of ubiquinone (coenzyme Q) in mammalian organisms. In addition to its well-established function as a component of the mitochondrial respiratory chain, ubiquinone has in recent years acquired increasing attention with regard to its function in the reduced form (...

Antioxidative effect of ubiquinones on mitochondrial membranes.

Peroxidation of mitochondria occurs extensively in ubiquinone-depleted membranes. Reincorporation into the membranes of either the physiological ubiquinone or a short-chain homologue protects mitochondria against peroxidation. The ability to prevent this phenomenon is more evident in mitochondria that have incorporated ubiquinone-3 and might be ascribed to an ordering structural effect on the l...
